Schematic of neutrophil PARP-1-ALOX5-mediated MMP-9 in lung cancer. Infiltrated neutrophils are increased in lung cancer tissues and negatively correlated with the prognosis of patients. After neutrophils are exposed to lung cancer cells, PARP-1 interacts with ALOX5 and enhances protein stabilization through PARylation of ALOX5. Increasing ALOX5 metabolites promotes MMP-9 production via activation of ERK and p38 MAPK pathways. Blocking PARP-1 by AG14361 or ALOX5 by Zileuton reduces MMP-9 production and mitigates neutrophil-mediated lung cancer progression. Credit: Cancer Biology & Medicine (2024).
